The herbal medicine is actively applied to the treatment of corona virus infectious diseases and pharmacological network is one of suitable tool for screening candidate drugs. We established a herb-compound-protein-disease pharmacological network for herbal medicines in the Korean, Chinese, and Japanese pharmacopeias. To analyze the built network in backward direction, we designed weight models of the main symptoms and sequelae of COVID-19 from previous studies. Then we explored the general efficacy of the identified herbs through compositional compounds, target proteins and literature reviews. Through backward analysis, Cichorii intybi radix, Glycine semen nigra, and Granati fructus are expected to have the greatest therapeutic efficacy for COVID-19 symptoms. Cnidii rhizoma, Foeniculi amari fructus, and Thymi herba showed potential therapeutic efficacy for COVID-19 sequelae. From literature reviews, we found that they have efficacy such as anti-inflammatory, anti-oxidant, anti-bacterial, anti-thrombotic, and immunomodulatory effects and contain some of the leading substances for COVID-19 treatments. We showed that pharmacological networks and backward network analysis could recommend herbal medicines expected to have efficacy for COVID-19 symptom and sequelae relief. The data-driven approach adopted in this work has shown the potential to function as a tool for selecting candidate herbal medicines and screening natural product-based leading substances. |
